Forensic Sibling Test
Discreet, Accurate DNA Testing Using Non-Standard Samples
A Forensic Sibling DNA Test helps determine whether two individuals share a biological sibling relationship when a standard cheek-swab sample is unavailable or increased discretion is needed.
Instead of requiring a traditional DNA collection, forensic sibling testing can use personal items and other non-standard samples that may contain DNA, allowing possible full- or half-sibling relationships to be investigated in situations where conventional testing may not be practical.

What Is a Forensic Sibling DNA Test?
A Forensic Sibling DNA Test combines DNA extraction from a non-standard sample with sibling relationship analysis.
The laboratory first attempts to recover sufficient DNA from the submitted item. When an interpretable DNA profile can be developed, that profile is compared with DNA from the other tested individual to evaluate the likelihood of a biological sibling relationship.
Depending on the relationship being investigated, testing can evaluate whether two individuals are likely to be:
Full siblings who share both biological parents
Half siblings who share one biological parent
Unlikely to share the proposed sibling relationship
Unlike direct paternity testing, sibling DNA testing is based on statistical relationship analysis. The laboratory evaluates shared genetic markers and calculates how strongly the DNA evidence supports the proposed relationship.
When Is a Forensic Sibling DNA Test Needed?
Forensic sibling DNA testing may be appropriate when:
One or both individuals cannot provide a standard cheek swab
Increased discretion is needed
Only a personal item from an individual is available
A possible sibling relationship needs to be investigated using an alternative DNA sample
A standard home Sibling DNA Test is not practical for the situation
This provides another testing option when conventional DNA collection is unavailable or inappropriate.
What Samples Can Be Used for Forensic Sibling DNA Testing?
Many personal items and biological materials may contain enough DNA for forensic testing.
Possible non-standard samples include:
Toothbrushes
Hair with roots attached
Fingernail or toenail clippings
Razors
Clothing or fabric
Other personal-use items that may contain biological material
Not every item will contain enough usable DNA for testing. The amount and quality of recoverable DNA can vary depending on the type of item, how it was used, its condition, storage, environmental exposure, and whether DNA from other individuals may also be present.
When possible, a toothbrush or another item likely to contain a single individual's DNA is generally preferable to an item that may have been handled or used by multiple people.
If you are unsure whether your item is suitable, our DNA experts can help you evaluate your available sample before ordering.

Step 2. Submit the DNA Samples
Submit the non-standard sample along with the DNA sample from the other participant.
The laboratory will attempt to recover DNA from the non-standard sample and determine whether sufficient genetic information is available for relationship testing.
Because non-standard samples vary considerably, DNA recovery cannot be guaranteed.
Step 3. Receive Your Results
When sufficient DNA is recovered, the developed DNA profiles are analyzed to evaluate the proposed sibling relationship.
Results are securely delivered by email and explain whether the genetic evidence supports the proposed relationship.
Understanding Your Forensic Sibling DNA Test Results
When sufficient DNA is recovered from the submitted samples, the sibling relationship analysis is performed using the same type of genetic and statistical principles used for sibling relationship testing with standard samples.
Results may indicate:
Relationship Supported
The genetic evidence supports the proposed sibling relationship.
Relationship Not Supported
The genetic evidence does not support the proposed sibling relationship.
Inconclusive
The available genetic evidence is not strong enough to clearly support or not support the proposed relationship.
An inconclusive sibling result does not automatically mean the individuals are unrelated. Depending on the circumstances, DNA from an additional known biological family member may provide additional genetic information and strengthen the analysis.

Non-Standard Samples and DNA Recovery
One of the most important differences between a home Sibling DNA Test and a Forensic Sibling DNA Test is the type of DNA sample being analyzed.
A cheek swab is collected specifically to provide a high-quality DNA sample. A non-standard sample was generally created or used for another purpose and may contain:
A strong single-source DNA profile
A limited or partial DNA profile
DNA from multiple individuals
Degraded DNA
Insufficient DNA for relationship testing
For this reason, successful DNA extraction from a non-standard sample cannot be guaranteed.
If sufficient DNA cannot be recovered, additional testing or another sample may be recommended.

Home or Forensic Sibling DNA Testing?
Both tests evaluate a possible biological sibling relationship. The primary difference is how the DNA sample is obtained.
Home Sibling DNA Test
Choose a Home Sibling DNA Test when both individuals can knowingly provide standard cheek-swab samples.
This is generally the simplest option for private, personal-knowledge sibling testing.

Forensic Sibling DNA Test
Choose a Forensic Sibling DNA Test when a standard cheek swab is unavailable or when testing needs to be performed using a personal item or another non-standard DNA sample.
Because DNA must first be recovered from the submitted item, forensic testing introduces an additional variable that is not normally present with standard cheek-swab testing.

Forensic Sibling DNA Testing:
Frequently Asked Questions
-
A Forensic Sibling DNA Test evaluates the likelihood that two individuals share a biological sibling relationship when a standard oral swab is not available from one or more participants. DNA may instead be recovered from an approved non-standard sample and used for sibling relationship analysis.
-
Yes. In some cases, a non-standard sample can be used in place of an oral cheek swab. Examples may include a toothbrush, razor, hair with roots, nail clippings, or other personal items that may contain sufficient DNA. DNA recovery from non-standard samples cannot be guaranteed.
Note that the cost will increase depending on the number of non-standard samples used in testing.
-
Yes. Sibling DNA testing can evaluate whether two individuals are more likely to be full siblings, half siblings, or unrelated. Because sibling testing is based on statistical relationship analysis rather than the direct inclusion or exclusion typically associated with paternity testing, the strength of the results can vary depending on the individuals tested and the DNA information available.
-
When a usable DNA profile is successfully recovered from the forensic sample, the laboratory compares the participants' DNA profiles and calculates statistical evidence supporting the tested sibling relationship. Including additional biological relatives when appropriate may provide additional genetic information and strengthen the analysis.
-
We strongly recommend verifying that a non-standard sample can produce a usable DNA profile when there is uncertainty about the sample.
Our DNA Profile Verification Test attempts to extract DNA and generate a DNA profile before proceeding with relationship testing, helping reduce the risk of an unsuccessful sibling analysis due to insufficient DNA.
-
DNA recovery from non-standard samples cannot be guaranteed. If sufficient usable DNA cannot be recovered, the sibling relationship analysis cannot be completed using that sample. Depending on the situation, another non-standard sample may be submitted or other testing options may be available.
